General description

Suppressor of fused homolog (UniProt: Q9UMX1; also known as SUFUH) is encoded by the SUFU (also known as UNQ650/PRO1280) gene (Gene ID: 51684) gene in human. SUFUH acts as a negative regulator in the hedgehog signaling pathway that is ubiquitously distributed in adult tissues. Three isoforms of SUFUH have been described that are produced by alternative splicing. Isoform 1 is detected in fetal brain, lung, kidney and testis and isoform 2 is detected in fetal testis, and at much lower levels in fetal brain, lung and kidney. SUFUH is shown to down-regulate Gli1-and Gli2-mediated transactivation of target genes. Binding of SUFUH sequesters Gli proteins in the cytoplasm. It is a part of a corepressor complex that acts on DNA-bound Gli1 and may link Gli1 to beta-transducin repeat containing E3 Ubiquitin Protein Ligase (BTRC) and facilitate its proteasomal degradation. SUFUH regulates the formation of either the repressor form (Gli3R) or the activator form (Gli3A) of the full length form of GLi3 (Gli3FL). When complexed with SUFUH, Gli3FL remains in the cytoplasm and is maintained in a neutral state. Mutations in SUFU gene are known to cause medulloblastoma.
